Titolo tesi: The Role of Chromosomal Instability in Regulating Long Non-Coding RNAs and Their Impact on Colorectal Cancer
Abstract: : This doctoral thesis investigates the impact of chromosomal instability (CIN) on long non-coding RNA (lncRNA) expression and evaluates their role in Colorectal Cancer (CRC) development, progression, and therapy. Chromosomal aberrations, such as gains in chromosomes 8 or 20, significantly influence the CRC transcriptomic profile, often upregulating oncogenic transcripts. Focusing on lncRNAs, known regulators of gene expression, this study aims to uncover their contribution to CRC pathology. The first phase will identify lncRNAs differentially expressed between tumor and healthy samples, correlating these changes with common chromosomal aberrations. Using SNP 6.0 arrays for chromosomal mapping and HTA 2.0 arrays for transcriptomic analysis, we will characterize the altered lncRNA expression profile in CRC tissues and cell lines. Subsequent phases will explore lncRNA mechanisms of action. Computational tools will predict their interactions with miRNAs and mRNAs, with experimental validation through techniques such as Dual Luciferase Reporter Assay. Correlation analysis will elucidate the regulatory RNA networks driving CRC oncogenesis. Additionally, the project will assess lncRNAs as prognostic biomarkers by correlating their expression with clinical outcomes, such as TNM staging, overall survival, and disease-free survival. Lastly, therapeutic potential will be explored by modulating lncRNA expression via siRNA and antisense oligonucleotides (ASOs) to examine their impact on cellular functions like proliferation, invasion, and apoptosis. This comprehensive study aims to elucidate the multifaceted role of lncRNAs in CRC, paving the way for novel biomarkers and personalized therapeutic strategies.
